Research Project

Quantification of Acylcarnitines in Patient’s Specimens with Organic Acidemias and Fatty Acid Oxidation Defects
Quantification of Medium- and Short-Chain Acyl-CoAs in Organic Samples by HPLC-MS/MS
Metabolic Toxicity of 6-Mercaptopurine and Azathiopurine

Simultaneous quantification of approximately 20 acylcarnitines (ACs) containing their isomers has been being developed by usage of HPLC-MS/MS (Figure 1). This method can be applied to the newborn screening for organic acidemias etc. AC profiles have been examined in biopsied and autopsied tissues of the patients
Medium- and short-chain acyl-CoAs in organic samples have been quantified for the elucidation of correlations among diseases, drugs and fatty acid metabolism.
Toxic metabolites of 6-mercaptopurine in human erythrocyte saline solution have been investigated.
Figure 1. LC-MS/MS spectra of ACs obtained by MRM mode: 1, carnitine (C); 2, malonyl-C; 3, acetyl-C; 4, succinyl-C; 5, glutaryl-C; 6, methylmalonyl-C; 7, propionyl-C; 8, isobutyryl-C; 9, butyryl-C; 10, pivaloyl-C; 11, 2-methylbutyryl-C; 12, isovaleryl-C; 13, valeryl-C; 14, hexanoyl-C; 15, heptanoyl-C; 16, valproyl-C; 17, octanoyl-C.
